Rezumat: Spectrofluorimetric measurements are useful tools to investigate structural changes in proteins. More insights into the protein folding pathways are essential for the general understanding of the antibody molecule structure. Moreover, elucidation of the folding mechanisms plays a crucial role in illustrating the final configurations of multidomain oligomeric proteins. The fact that chicken IgY antibodies might represent exquisite tools in diagnostics, medical application or biotechnology prompted us to investigate the effect of chemical conditions- urea treatment on IgY stability, by monitoring changes in fluorescence emission spectra. Our data revealed that IgY stability is not affected after 3-5 hours incubation with 0-9M urea, denaturation occuring only after 21h incubation with 5,5-9.5M urea. The low fluorescence intensity of ANS bound to IgY, appears to indicate no conformational changes after urea treatment, probably due to the lack of hydrophobic sites on IgY immunoglobulin. Increase stability in different urea conditions would suggest a possible therapeutic role of IgY antibodies. | |||||||
