Abstract:  From a survey of the diversity of Bacillus-like species, a novel strain FJAT-45037 was found to be differing from the known Bacillus species. In this study, the taxonomic status of this potential new species was determined using genomics-based polyphasic taxonomy (Ramasamy et al., 2014). Cells of strain FJAT-45037 were aerobic, endospore-forming and motile. Temperature range for growth was 10 to 35 o C (optimum 30 o C). pH range for growth was 7.5~12.0 (optimum 9.0~10.0). NaCl concentration range for growth was 0-12% (w/v) (optimum 1%). The 16S rRNA gene sequence-based phylogenetic analysis revealed that strain FJAT-45037 is most closely related to Bacillus marmarensis GMBE 72T (97.67%), and their DNA-DNA hybridization (isDDH) value was 18.6% with an average nucleotide identity (ANI) value of 84.46%. The DNA G+C content was 39.79 mol%. The major cellular fatty acids were anteiso-C15:0 and anteiso-C17:0. The diagnostic diamino acid of the cell-wall peptidoglycan was meso-diaminopimelic acid. MK-6 and MK-7 were the predominant quinones. Major polar lipids were diphosphatidyl glycerol, phosphatidyl glycerol and phosphatidyl ethanolamine. On the basis of the polyphasic evidence presented, the strain was considered to represent a novel member of the genus Bacillus, for which the name Bacillus alkalisalsus sp. nov. was proposed. The type strain is FJAT-45037 (= DSM 104053 = CCTCC AB 2016229).

Keywords: alkalitolerant, Bacillus alkalisalsus, genome, polyphasic taxonomy.
